IL-23R (Interleukin 23 Receptor) encodes a subunit of the receptor for interleukin-23 (IL-23), a pro-inflammatory cytokine that plays a crucial role in the immune response. The IL-23 receptor is expressed on various immune cells including T helper 17 (Th17) cells, natural killer cells, and macrophages.IL-23 signaling through its receptor promotes the differentiation and maintenance of Th17 cells, which produce pro-inflammatory cytokines such as IL-17, IL-21, and IL-22. This pathway is essential for host defense against extracellular bacteria and fungi, but dysregulated IL-23/IL-23R signaling has been implicated in the pathogenesis of various autoimmune and inflammatory disorders.
Genetic variants in IL-23R have been strongly associated with susceptibility to inflammatory bowel disease (both Crohn's disease and ulcerative colitis), psoriasis, ankylosing spondylitis, and other immune-mediated conditions. Interestingly, some IL-23R variants confer protection against these diseases, making this gene an important target for understanding immune regulation and developing targeted therapies.
